This fitting connects to PEX via the crimp method. Uponor currently does not offer a ProPEX drop ear elbow with 3/4" PEX and 1/2" NPT connections.
I have seen them come either way. It depends on the manufacturer.

I don't remember exactly, but I think its three. What I do remember is that they felt very secure after installation. I didn't have any concerns.
If I recall correctly, I have used these elbows with both two and three screws. The top screw that you are referring to, that may or may not be there is insignificant because the pipe is coming up preventing much less pressure that would be needed by that screw. If you get the two or the three screw elbow, both will work fine.
The ones I received had three screw holes.
You couldn't, because the MANABLOC has unique threads that do not work with standard pipe thread fittings. Only MANABLOC fittings are compatible.
